Best to pull old tube out before putting new one in.

Be sure to pull plug, also.

Seriously, the effort you put in to replace the tube with an air cooled tube
could be put into finding a used replacement.  If you are determined, you
could use the conduction cooled tetrode version of the 4CX250 and then
install a screen regulator circuit and continue to drive cathode, ground
control grid for rf, bias tube, and use screen voltage.

You could also take the time invested, give the amp away to a charitable
organization, work for $10 an hour sweeping out horse stalls and buy a new
amp.
