Yes.
Select 2 for Number of Radios in the Radio menu. This will create two Entry
windows.
In the Setup menu select Ports and setup two instances of the K3 on the same
COM port where your K3 is connected.
In the Radio button in the upper Entry window, click the drop-down menu in
Log the frequency of QSOs, and select the Comm port entry.
In the Radio button in the lower Entry window, do the same thing but select
Comm port Rig 2.
When the TX focus is on the lower (sub-RX) Entry window, WriteLog puts the
K3 in SPLIT mode so you can transmit there. For RTTY FSK, this means you'll
need two FSK keying interfaces, each on a different Com port and each of
those Com ports associated with different Rttyrite windows with MMTTY or
whatever software modem you use. Same thing for hardware TNCs. For CW, if
you use the K3 COM port keying on the DTR (or, RTS) signal, then you can
just tick the CW button in the Ports menu for both K3 entries. If a WinKey
or MicroHAM box or other keyer, just specify its COM port as a third COMn
entry in the Ports window.
