We have been fortunate to contact a descendant of John Hooper, Jr., William Alsup and Samuel Perrin (Spelled Sperring in St. Paul's Vestry Records). We will quote some brief records and if anyone is interested in further information, we will put you in touch with David Alsup.
"Based on my findings, John Hooper was in Hanover County. Co. during mid-1700's. He was living with my Alsup family (per St. Paul's' Parish documents) and he was processioned with land holdings in the same district as my Alsup family.

According to Hooper Genealogy, A Southern Branch by Flora Hooper Collier, 1972, Pg. 4 "There was a John Hooper as early as Sept. 16, 1710. The first entry for him was for "Doctor and medicine for ". October 5, 1731, an entry "for keeping him". This would indicate that he was a very old man, and may have died after this entry, for Oct. 10, 1744 there was an entry for a John Hooper who was "paid for keeping Joseph Sperring ". This would indicate a younger man. Sept. 30, 1751 was the last date for John Hooper.
We have no information on any other children for the first John Hooper.
John Hooper, James Hooper, William Hooper and Edward Hooper were found in Hanover County 1743/44.
