Yes.

Most contest software that I am aware of will not allow 
simultaneous transmission on more than one radio.  The
computer can only key one radio at a time. Some ops use 
two computers for SO2R.  In such cases, they must build
a lockout device to prevent simultaneous transmission.

Receive audio switching is left up to the imagination,
but only one transmitted signal is permitted for Single Op.
