How to measure?

Connect a 50 ohm dummy load in place of one of the antenna ports.

Insert a wattmeter at the amplifier output.
Tune up on 28 MHz.
Measure power output (P1).

Move the same wattmeter to the 50 ohm load.
Measure power output  (P2).

Compare.

Loss =  10 LOG P2 / P1.

Note that your wattmeter accuracy is greatest near full scale
therefore tuning to 200 Watts on a 200 Watt scale is a good
way to make your measurement without cooking your load
at full power.
