Agreed, except for the tune cap, I would leave it connected to the
tube anode, so the tube's capacitance is included in your
measurements. After all, that is the actual value you will be using in
the amp. But you can add it in later if you want. Your call. 

If you do have the tube connected and you are using a low frequency
C-meter such as the AADE unit, disconnect the plate choke or it will
throw you off, way off.
